Background:

  • Preeclampsia is a serious pregnancy complication characterized by high blood pressure and potential organ damage.
  • Nitric oxide (NO) plays a crucial role in regulating vascular tone and blood pressure during pregnancy.

Purpose of the Study:

  • To investigate and clarify the production of nitric oxide in women with preeclampsia compared to healthy pregnant women.
  • To assess the relationship between nitric oxide metabolites and other endothelial dysfunction markers in preeclampsia.

Main Methods:

  • Simultaneous measurement of plasma nitrite and nitrate levels and renal clearance in 20 preeclamptic and 21 healthy pregnant women.
  • Subjects adhered to low-nitrite/nitrate diets and fasted overnight prior to sample collection.

  • Nitrite and nitrate concentrations were determined using the spectrophotometric Griess reaction.

    • Preeclamptic women exhibited significantly higher plasma nitrite and nitrate levels (18.1±6.2 vs. 13.0±4.3 µmol/L) compared to controls.
    • Renal clearance of nitrite and nitrate did not differ between groups, suggesting increased NO production in preeclampsia.
    • Elevated endothelin-1 and decreased 2,3-dinor-6-keto-prostaglandin F1α were observed in preeclampsia, but without correlation to NO metabolites.

    Conclusions:

    • Nitric oxide production is significantly increased in women with preeclampsia.
    • The biological significance of this enhanced nitric oxide production remains unclear but may serve as a compensatory mechanism against vasoconstriction.
    • Findings were independent of antihypertensive medications or betamethasone treatment received by some participants.
