Summary

T lymphocytes from systemic sclerosis (SSc) patients can induce tissue factor (TF) expression in endothelial cells. However, this induction was similar in healthy controls, suggesting T cells are not uniquely responsible for TF changes in SSc.

Background:

  • Systemic sclerosis (SSc) is an autoimmune disease characterized by vascular damage.
  • Tissue factor (TF) plays a critical role in thrombosis and inflammation.
  • The role of T lymphocytes in TF induction in SSc is not fully understood.

Purpose of the Study:

  • To investigate the capacity of circulating T lymphocytes from SSc patients to induce TF expression in human umbilical vein endothelial cells (HUVECs).
  • To compare TF induction by T lymphocytes from SSc patients with that from healthy controls.

Main Methods:

  • Peripheral blood T lymphocytes were isolated from nine SSc patients and ten healthy controls.
  • T lymphocytes were co-cultured with HUVECs at varying concentrations.

  • Main Results:

    • T lymphocytes from SSc patients induced TF activity in HUVECs in a dose-dependent manner.
    • Significant TF induction was observed at lymphocyte concentrations of 1x10^6 and 2x10^6 cells/well.
    • No significant differences in endothelial TF induction were found between SSc patients and healthy controls across tested lymphocyte concentrations.

    Conclusions:

    • Circulating T lymphocytes can induce TF expression in endothelial cells, but this capability is not unique to SSc patients.
    • The study did not find a correlation between TF activity induced by T cells and clinical features or disease duration in SSc patients.
