Summary

Alprazolam, a GABA agonist, increased dehydroepiandrosterone (DHEA) levels and decreased cortisol in healthy men. DHEA concentrations were U-shapedly related to psychomotor impairment after alprazolam.

Area of Science:

  • Neuroendocrinology
  • Pharmacology

Background:

  • Alprazolam (a gamma-aminobutyric acid agonist) reduces cortisol and adrenocorticotropic hormone.
  • Dehydroepiandrosterone (DHEA) and its metabolite DHEA-S may antagonize GABA activity.
  • DHEA and DHEA-S exhibit diurnal variations and are secreted with cortisol.

Purpose of the Study:

  • To investigate alprazolam's effect on DHEA and DHEA-S concentrations.
  • To explore the relationship between DHEA/DHEA-S levels and alprazolam-induced psychomotor impairment.

Main Methods:

  • Healthy young and elderly men received intravenous alprazolam.
  • Plasma steroid concentrations (DHEA, DHEA-S, cortisol) were measured over 9 hours.
  • Psychomotor function was assessed in relation to steroid levels.

Related Experiment Videos

Main Results:

  • Alprazolam significantly increased DHEA concentrations at 7 hours.
  • Cortisol levels decreased significantly after alprazolam administration.
  • No significant changes in DHEA-S concentrations were observed.
  • A U-shaped relationship was found between DHEA levels and psychomotor decrement.

Conclusions:

  • Alprazolam modulates peripheral DHEA concentrations.
  • DHEA and/or DHEA-S may play a role in vivo in GABA receptor-mediated responses.
  • DHEA levels are linked to psychomotor impairment following alprazolam exposure.
