The COBE Spectra Version 6.0 automated system for peripheral blood progenitor cell (PBPC) collection showed comparable results to Version 4.7, except for lower CD34+ cell counts where Version 4.7 was superior. Version 6.0 significantly reduced platelet contamination in autografts.

Area of Science:

  • Hematology
  • Biomedical Engineering
  • Cellular Therapy

Background:

  • Peripheral blood progenitor cell (PBPC) collection traditionally involved semi-automated processes requiring manual operator intervention.
  • The COBE Spectra apheresis device, with its Version 6.0 software, introduced an automated program for PBPC collection.

Purpose of the Study:

  • To compare the efficacy and product quality of PBPC collection using COBE Spectra Version 6.0 against Version 4.7.
  • To evaluate the impact of software versions on CD34+ cell yield and product purity.

Main Methods:

  • A comparative study involving 46 patients undergoing PBPC collection.
  • Patients were randomized to processing with either COBE Spectra Version 4.7 (n=24) or Version 6.0 (n=22).
  • Analysis focused on CD34+ cell count, mononuclear cell (MNC) count, white cell count (WCC), hemoglobin (Hb), and platelet content in the autograft, stratified by baseline PB CD34+ cell counts.

Related Experiment Videos

Main Results:

  • No significant differences in autograft product quality were observed between Version 4.7 and 6.0 when baseline peripheral blood CD34+ cell counts were 10-50x10(6)/L or >50x10(6)/L.
  • Version 4.7 demonstrated superior performance in collecting CD34+ cells at lower starting counts (<10x10(6)/L).
  • Autograft platelet contamination was significantly lower with Version 6.0 compared to Version 4.7 (P<0.0001).

Conclusions:

  • COBE Spectra Version 6.0 offers comparable PBPC collection efficiency to Version 4.7 across most CD34+ cell count ranges.
  • Version 6.0 provides a significant advantage by reducing platelet contamination in the autograft product.
  • The choice of software version may depend on the patient's baseline CD34+ cell count and the desired product purity.
