    Summary

    Postnatal lead exposure in rats did not cause hyperactivity. This study suggests that low-level lead exposure may not reliably induce hyperactivity in rodents, challenging previous findings.

    Background:

    • Previous research suggested postnatal lead exposure causes hyperactivity in rodents.
    • A rodent model for childhood hyperactivity has been proposed based on lead exposure.

    Purpose of the Study:

    • To investigate the effects of postnatal lead exposure on rodent behavior.
    • To assess the validity of lead-induced hyperactivity as a model for childhood hyperactivity.

    Main Methods:

    • Offspring of lead-intubated and pair-fed control mothers were used.
    • Locomotor activity was measured in rats aged 24-27 days.
    • Behavioral observations were conducted at 28-29 and 35-36 days of age.
    • Brain lead concentrations were analyzed.

    Main Results:

    • No significant differences in locomotor activity or basal activity were observed between lead-exposed and control rats.
    • Active behaviors were slightly reduced in lead-exposed rats.
    • Brain lead concentrations were significantly elevated in lead-exposed rats.
    • Statistical power analyses indicated that previously reported hyperactivity effects would likely have been detected.

    Conclusions:

    • Low-level postnatal lead exposure may not reliably produce hyperactivity in rodents.
    • Existing data offer limited support for lead-induced hyperactivity in rodents as a model for childhood hyperactivity.
