Consuming chargrilled meat induces CYP1A enzymes in the liver and small intestine, but not CYP3A4 or P-glycoprotein. This suggests an adaptive role for CYP1A in response to carcinogens in chargrilled meat.
Area of Science:
- Biochemistry
- Toxicology
- Gastroenterology
Background:
- Chargrilled meat contains carcinogens like heterocyclic amines and polycyclic aromatic hydrocarbons.
- These compounds are substrates for inducible CYP1A and CYP3A enzymes and P-glycoprotein.
- The study investigates the effect of chargrilled meat consumption on these proteins.
Purpose of the Study:
- To determine if a diet high in chargrilled meat induces CYP1A, CYP3A enzymes, and P-glycoprotein.
- To assess the adaptive response of these proteins to dietary carcinogens.
Main Methods:
- Ten healthy adults consumed a chargrilled meat-enriched diet for 7 days.
- Biopsies of the small intestine and colon were analyzed for CYP1A, CYP3A, and P-glycoprotein mRNA and protein.
- Hepatic enzyme activity and DNA adduct levels were measured.
Main Results:
- Chargrilled meat consumption unequivocally induced CYP1A enzymes in the liver and small intestine.
- No induction of CYP3A4, CYP3A5, or P-glycoprotein was observed in the small intestine or colon.
- An inverse correlation was found between polycyclic aromatic hydrocarbon DNA adducts and CYP1A activity/protein levels.
Conclusions:
- Dietary chargrilled meat induces CYP1A enzymes, suggesting an adaptive role.
- CYP3A4 and P-glycoprotein were not induced by chargrilled meat consumption.
- The findings support CYP1A's role in adapting to carcinogens found in chargrilled meat.
