Background:

  • Growth hormone receptors (GHRs) and prolactin receptors (PRLRs) are crucial for mediating the effects of growth hormone (GH) and prolactin (PRL).
  • Understanding the expression patterns of these receptors in peripheral blood mononuclear cells (PBMC) is important for insights into immune function and endocrine regulation.
  • Previous studies have not fully elucidated the age- and sex-related variations in GHR and PRLR expression on human PBMC subsets.

Purpose of the Study:

  • To investigate the expression levels of GHR and PRLR in human PBMC.
  • To examine how GHR and PRLR expression varies across different age groups and in relation to sex.
  • To explore the correlation between plasma GH binding protein (GHBP) and GH receptor expression on PBMC.

Main Methods:

  • Flow cytometry was employed to quantify GHR and PRLR expression on human PBMC.
  • Biotinylated anti-GHR monoclonal antibody 10B8 and biotinylated human PRL were used for receptor detection.
  • Double immunofluorescence staining allowed for the analysis of receptor expression on specific PBMC subsets (T cells, B cells, monocytes).

Main Results:

  • Approximately 30% of total PBMC expressed GH receptors, with low expression on T cells and high expression on B cells and monocytes.
  • The percentage of PBMC expressing GH receptors was significantly lower in the 20-40 year age group compared to younger (12-20 yr) and older (>60 yr) groups.
  • PRLR levels were significantly higher in the 20-40 year age group compared to the >60 year group, and a negative correlation was found between plasma GHBP and PBMC GHR percentage.

Conclusions:

  • GHR and PRLR expression on human PBMC exhibits significant age-dependent variations.
  • The regulation of GH receptors may differ between lymphocytes and other target cells.
  • These findings highlight the dynamic nature of hormone receptor expression in immune cells throughout the lifespan.
