Transforming growth factor beta 2 (TGF-beta 2) is highly expressed in fetal and glaucomatous optic nerve heads, suggesting its role in development and glaucoma pathogenesis. This study investigated TGF-beta isoforms in optic nerve heads.

Background:

Purpose of the Study:

  • To investigate the presence and localization of TGF-beta isoforms (TGF-beta 1, TGF-beta 2, TGF-beta 3) in human fetal, normal adult, and glaucomatous optic nerve heads.
  • To determine if TGF-beta synthesis and gene expression differ across these conditions.

  • Immunohistochemistry was used to localize TGF-beta isoforms in optic nerve head tissues.

    • Glaucomatous and fetal optic nerve heads released significantly higher levels of TGF-beta 2 compared to normal adult tissues.
    • TGF-beta 1 was undetectable by bioassay in all tested samples.
    • No significant increase in TGF-beta 2 gene expression was observed, suggesting post-transcriptional regulation.

    Conclusions:

    • TGF-beta 2 is abundantly produced in fetal and glaucomatous optic nerve heads, likely through post-transcriptional regulation.
    • TGF-beta 2 may play a role in optic nerve head development.
    • In glaucoma, TGF-beta 2 could mediate astrocyte activation and extracellular matrix remodeling in the lamina cribrosa.
