Summary

Human telomeres replicate throughout S phase, unlike yeast telomeres which replicate late. Telomere processing and replication timing in humans differ significantly from yeast, challenging previous hypotheses.

Area of Science:

  • Cell Biology
  • Genetics
  • Molecular Biology

Background:

  • Yeast telomeres replicate late and exhibit positional effects, suggesting heterochromatin formation and subtelomeric gene regulation.
  • Telomere replication and processing in yeast involve a transient long overhang at the end of S phase.

Purpose of the Study:

  • To investigate the timing of telomere replication and processing in normal human diploid fibroblasts.
  • To compare human telomere dynamics with those observed in yeast.

Main Methods:

  • Synchronized human fibroblasts were labeled with 5-bromodeoxyuridine (BrdU) at hourly intervals.
  • Telomeres were purified and analyzed for BrdU incorporation using anti-BrdU antibodies.
  • Telomere length and telomerase activity were assessed in relation to replication timing.

Related Experiment Videos

Main Results:

  • Human telomeres replicate throughout the S phase, not late as in yeast.
  • Replication timing was independent of telomere length, cell age, or telomerase-induced elongation.
  • Asymmetric G-rich overhangs were established within 1 hour of replication, indicating rapid processing.

Conclusions:

  • Human telomere replication and processing timings are distinct from yeast.
  • The hypothesis that human telomeres are heterochromatic and regulate subtelomeric genes based on yeast models requires re-evaluation.
