Background:

  • Protoporphyrin IX (PROTO) is a photosensitizing porphyrin implicated in certain toxicities.
  • Understanding the oxidative stress induced by PROTO is crucial for assessing its hepatic effects.

Purpose of the Study:

  • To investigate the short- and long-term pro-oxidant effects of PROTO administration on mouse liver.
  • To characterize the impact of PROTO on oxidative stress markers and antioxidant enzyme activities in the liver.

Main Methods:

  • Mice were administered PROTO (3.5 mg/kg, i.p.) to assess liver porphyrin accumulation.
  • Assays were performed to measure delta-aminolevulinic acid synthetase activity, lipid peroxidation (TBARS), glutathione (GSH) content, and activities of antioxidant enzymes (SOD, catalase, GPx, GRed, GST).

Main Results:

  • PROTO administration led to rapid liver porphyrin accumulation and induced delta-aminolevulinic acid synthetase activity.
  • Initial PROTO exposure triggered oxidative stress, increasing antioxidant enzyme activities (SOD, catalase, GPx, GRed) to combat lipid peroxidation and GSH depletion.
  • Sustained PROTO accumulation resulted in severe liver damage, compromising antioxidant defenses with reduced SOD, GPx, and catalase activities, and diminished GST activity.

Conclusions:

  • PROTO accumulation in the liver induces significant oxidative stress and initiates a compensatory increase in antioxidant enzyme activity.
  • Chronic exposure and severe liver damage due to sustained PROTO accumulation overwhelm the hepatic antioxidant system, leading to compromised protection.
