Area of Science:

  • Genetics
  • Toxicology
  • Molecular Biology

Background:

  • The MutaMouse system is widely used for in vivo somatic mutation quantification.
  • The lacZ gene, commonly used in MutaMouse, presents challenges for efficient sequence analysis due to its large size.
  • A need exists for alternative, more easily analyzed mutational targets.

Purpose of the Study:

  • To report the first application and characterization of the cII locus as a mutational target in the MutaMouse system.
  • To compare the performance of the cII locus with the lacZ gene in terms of spontaneous and induced mutation frequencies.
  • To evaluate the suitability of the cII locus for efficient sequence spectra analysis and jackpot mutation identification.

Main Methods:

  • Application and characterization of the cII locus in the MutaMouse system.

Related Experiment Videos

  • Treatment of MutaMice with N-ethyl-N-nitrosourea (ENU) at various doses.
  • Comparison of mutation frequencies at cII and lacZ loci.
  • Main Results:

    • The cII locus demonstrated similar spontaneous and induced mutation frequencies compared to the lacZ gene.
    • Mutant frequencies at the cII locus reached a maximum 10 days post-ENU treatment and remained constant.
    • The cII locus showed a linear dose-response to ENU treatment.
    • The smaller size of the cII locus (0.3 kb) facilitates efficient sequence spectra analysis compared to the lacZ gene (3.0 kb).

    Conclusions:

    • The cII locus is a genetically neutral and effective mutational target for the MutaMouse system.
    • The cII locus offers improved efficiency for sequence analysis and jackpot mutation identification.
    • The cII locus provides a common transgenic target for both MutaMouse and Big Blue in vivo mutagenesis assays.
