    Background:

    • Cholestyramine is a widely used medication for lowering cholesterol.
    • Traditional administration involves multiple daily doses (t.i.d.), which can be inconvenient for patients.

    Purpose of the Study:

    • To evaluate a simplified, once-daily (s.i.d.) administration method for cholestyramine.
    • To compare the efficacy of s.i.d. cholestyramine dosing with traditional t.i.d. dosing in lowering serum cholesterol.

    Main Methods:

    • Adult patients received cholestyramine in single daily doses (12-16 gm) or divided twice-daily doses (b.i.d.) for larger doses (24 gm).
    • Serum cholesterol reductions were compared between s.i.d., b.i.d., and historical t.i.d. regimens.
    • Serum triglyceride levels were monitored during therapy.

    Main Results:

    • Single daily doses of cholestyramine achieved cholesterol reductions comparable to or greater than traditional t.i.d. administration (21.58% vs. 25.61% average reduction).
    • A twice-daily (b.i.d.) regimen resulted in a 27.57% average cholesterol reduction.
    • Out-patients on the conventional t.i.d. program showed a 16.8% reduction, while the overall series of 238 patients averaged a 22% reduction.

    Conclusions:

    • Once-daily cholestyramine administration is a simplified and effective method for cholesterol reduction.
    • The simplified regimen maintains efficacy while potentially improving patient compliance.
    • Monitoring of serum triglycerides is recommended during cholestyramine therapy.
