    Summary

    Sera from children with acute lymphoblastic leukemia (ALL) can inhibit normal lymphocyte transformation. This effect was most pronounced in newly diagnosed patients with high white blood cell counts, suggesting a potential prognostic marker.

    Background:

    • Pokeweed mitogen (PWM) is a common stimulant for lymphocyte transformation.
    • Acute lymphoblastic leukemia (ALL) is a significant pediatric cancer requiring effective diagnostic and prognostic tools.

    Purpose of the Study:

    • To investigate the effect of sera from children with ALL on the in vitro transformation of normal lymphocytes.
    • To determine if specific patient characteristics correlate with inhibitory serum activity.

    Main Methods:

    • Normal lymphocyte cultures were stimulated with PWM.
    • Leukemic sera were added to lymphocyte cultures at varying concentrations (20% and 60%).
    • Lymphocyte transformation was assessed in the presence of sera.

    • Sera from children with ALL significantly inhibited PWM-induced lymphocyte transformation.
    • Inhibitory activity was primarily observed in pretreatment sera from newly diagnosed ALL patients with leukocyte counts >50,000/mm3.

    Conclusions:

    • A serum inhibitor of lymphocyte transformation exists in a subset of ALL patients.
    • This inhibitor may be associated with high tumor burden at diagnosis.
    • Further research is needed to explore the clinical significance of this inhibitor for patient prognosis.
