Summary

A new colorimetric assay accurately detects alditols, like mannitol, in biological samples. This rapid, cost-effective method uses simple reagents and achieves nanomolar sensitivity with minimal interference.

Area of Science:

  • Biochemistry
  • Analytical Chemistry

Background:

  • Alditol quantification is crucial for understanding various biological processes.
  • Existing methods for alditol determination can be complex, costly, or prone to interference.

Purpose of the Study:

  • To develop a simple, rapid, and sensitive colorimetric assay for alditol determination.
  • To minimize interference from other carbohydrates in biological extracts.

Main Methods:

  • Alditol oxidation using sodium periodate under acidic conditions.
  • Detection of formaldehyde byproduct via Hantzsch reaction with ammonia and acetylacetone.
  • Spectrophotometric measurement of the resulting colored heterocycle.

Main Results:

  • The assay demonstrates negligible interference from other carbohydrates.
