Synergism is a useful mechanism where combining two or more drugs is more effective than each constituent used alone. Such combinations are also called supra-additive interactions. The drugs collectively enhance the final therapeutic effect by acting on different targets. Adding etanercept to methotrexate significantly improved outcomes for rheumatoid arthritis patients with persistent disease activity. This combination therapy demonstrated superior efficacy compared to methotrexate alone.

  • Methotrexate (MTX) is a common monotherapy for rheumatoid arthritis (RA).

Purpose of the Study:

  • To evaluate the efficacy of combining etanercept with methotrexate (MTX) in RA patients.
  • To assess treatment outcomes in patients with persistent RA activity despite MTX monotherapy.

Main Methods:

  • A randomized controlled trial comparing etanercept plus MTX versus placebo plus MTX.
  • Patients had active RA despite stable MTX monotherapy.
  • Efficacy was assessed using American College of Rheumatology (ACR) response criteria at 6 months.
