Nucleotide sequence of microvariant RNA: another small replicating molecule.

D R Mills, F R Kramer, C Dobkin

Proceedings of the National Academy of Sciences of the United States of America
    |November 1, 1975

    Summary

Researchers discovered microvariant RNA, a small self-replicating molecule, from Qbeta replicase reactions. Its sequence similarity to MDV-1 RNA was negligible, suggesting structure, not sequence, dictates replication.

    Area of Science:

    • Molecular Biology
    • Virology
    • Biochemistry

    Background:

    • Qbeta replicase synthesizes RNA molecules.
    • Endogenous RNA products like MDV-1 RNA have been previously characterized.
    • Self-replicating RNA molecules can arise without external templates.

    Purpose of the Study:

    • To determine the nucleotide sequence of microvariant RNA.
    • To compare the sequence of microvariant RNA with MDV-1 RNA.
    • To investigate the factors governing Qbeta replicase recognition and RNA replication.

    Main Methods:

    • Isolation of microvariant RNA from Qbeta replicase reactions.
    • Determination of the complete nucleotide sequence of microvariant RNA.
    • Comparative sequence analysis between microvariant RNA and MDV-1 RNA.

    Related Experiment Videos

    Main Results:

    • Microvariant RNA (114 nucleotides) was isolated and sequenced.
    • No significant sequence similarity was found between microvariant RNA and MDV-1 RNA (220 nucleotides).
    • Qbeta replicase can synthesize both disparate RNA molecules, indicating sequence alone is insufficient for recognition.

    Conclusions:

    • Primary nucleotide sequence is not the sole determinant for Qbeta replicase recognition and RNA replication.
    • Secondary or tertiary structures are likely critical for enzyme recognition and replication.
    • The availability of two distinct replicating RNA molecules aids in identifying key structural features.
