Insights

Tumor necrosis factor (TNF) and oxidative stress increase superoxide dismutase (SOD) activity in melanoma and endothelial cells. This enhanced SOD activity appears to reduce the upregulation of intercellular adhesion molecule-1 (ICAM-1) expression.

Area of Science:

  • Biochemistry
  • Cell Biology
  • Immunology

Background:

  • Intercellular adhesion molecule-1 (ICAM-1) expression is upregulated by tumor necrosis factor (TNF) and oxidative stress.
  • Reactive oxygen species, including superoxide anion, activate transcription factors like NF-kappaB and AP-1, influencing ICAM-1.
  • Superoxide dismutase (SOD) enzymes (Cu/Zn-SOD and Mn-SOD) detoxify superoxide anions.

Purpose of the Study:

  • To investigate the role of superoxide anion generation and SOD activity in regulating ICAM-1 expression.
  • To examine the effects of TNF and oxidative stress on SOD activity and ICAM-1 levels in human melanoma and endothelial cells.

Main Methods:

  • Cells were exposed to TNF or Paraquat (a superoxide generator).
  • SOD activity was assessed using zymography, and protein levels via Western blotting and ELISA.
  • ICAM-1 expression was quantified; effects of SOD inhibition (DTIC) and a SOD mimetic (MnTMPyP) were evaluated.

Main Results:

  • TNF or Paraquat exposure increased both Cu/Zn-SOD and Mn-SOD activities (20-50%) and protein levels.
  • ICAM-1 expression significantly increased (140-200%) under TNF or Paraquat treatment.
  • Inhibition of Cu/Zn-SOD enhanced ICAM-1 upregulation, while a SOD mimetic reduced ICAM-1 expression, particularly under Paraquat stress.

Conclusions:

  • Melanoma and endothelial cells increase Mn- and Cu/Zn-SOD activities upon exposure to TNF or oxidative stress.
  • Elevated SOD activity is associated with reduced stimulation of ICAM-1 expression by TNF or oxidative stress.
  • These findings highlight a protective role of SOD in modulating inflammatory responses mediated by ICAM-1.

Immunoglobulin-like Cell Adhesion Molecules

Immunoglobulin-like cell adhesion molecules or Ig-CAMs are a versatile group of cell surface glycoproteins belonging to the immunoglobulin protein superfamily. Ig-CAMs possess the characteristic immunoglobulin protein domains and other domains such as the fibronectin type III domain. The Ig domains are glycosylated to varying degrees in different Ig-CAMs.
Ig-CAMs exhibit either homophilic binding (to other Ig-CAMs) or heterophilic binding (to other ligands such as integrins). While most Ig-CAMs...
Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl hydroxylase and factor...

Cancer Cell Migration through Invadopodia

Invadosome is a broad category of cell surface structures with proteolytic activity that  degrades the extracellular matrix (ECM). Invadosomes are present in normal cell types, including macrophages, endothelial cells, and neurons, as well as tumor cells. Although the macrophage podosomes and tumor cell invadopodia are classified as invadosomes, they have different structures, molecular pathways, and functions. Podosomes are short structures that last for a few minutes. However, invadopodia can...
Activation of Integrins01:15

Activation of Integrins

Integrins bind ligands and transmit information from outside the cell to inside or vice-versa through an "outside-in signaling" or "inside-out signaling."
In "outside-in signaling," external factors in the extracellular space bind to exposed ligand binding sites on integrins. This causes the inactive protein to undergo a conformational change to become active. Integrins are often clustered on the cell membrane. Repetitive and regularly spaced ligand binding events provide an effective stimulus.
Role Of Notch Signalling In Intestinal Stem Cell Renewal01:12

Role Of Notch Signalling In Intestinal Stem Cell Renewal

Notch signaling was first discovered in Drosophila melanogaster, where it is involved in cell lineage differentiation. Notch signaling regulates the maintenance and differentiation of intestinal stem cells or ISCs by controlling the expression of atonal homolog 1 or Atoh1. Atoh1 directs cells to differentiate into secretory cells.
Direct cell-to-cell contact is needed for the activation of Notch signaling. The signal is initiated when a notch ligand binds to a receptor on an adjacent cell, also...

Cell Adhesion Molecules - Types and Functions

Cell adhesion molecules (CAMs) are pivotal to multicellularity and the coordinated functioning of tissues and organ systems. They enable physical interactions between cells and provide mechanical strength to tissues. They also function as receptors for signal transmission across the plasma membrane. The CAMs are broadly classified into four families - integrins, cadherins, selectins, and immunoglobulin-like CAMs (IgCAMs).
