Summary

Inflammatory cytokines like interleukin-1 (IL-1) and tumor necrosis factor (TNF-alpha) directly impair gallbladder epithelial cell function. This disruption of absorptive processes may contribute to gallstone formation.

Background:

  • Gallbladder inflammation is an early indicator of gallstone development in animal models.
  • Inflammation involves increased myeloperoxidase and interleukin (IL)-1 activity in the gallbladder wall.

Purpose of the Study:

  • To investigate if inflammatory cytokines directly impact the absorptive function of gallbladder epithelial cells.
  • To understand the role of altered absorptive function in gallstone pathogenesis.

Main Methods:

  • Cultured human gallbladder epithelial cells were exposed to various agents, including interleukin-1 alpha (IL-1alpha), tumor necrosis factor alpha (TNF-alpha), lipopolysaccharide, and prostaglandin E2.
  • Unidirectional sodium and chloride fluxes were measured to calculate net ion transport.

    • Altered gallbladder absorptive function due to inflammation may play a critical role in the development of gallstones.
