Receptor Downregulation in MVBs01:15

Receptor Downregulation in MVBs

Multivesicular bodies (MVBs) are mature endosomes that sort ubiquitinated proteins and then fuse with lysosomes to degrade the sorted proteins. Epidermal growth factor (EGF) and its receptor (EGFR) form a complex that can be internalized through endocytosis, sorted into an MVB, and later degraded.
The EGFR can initiate signaling pathways that  lead to cell proliferation, migration, and differentiation. Overexpression of EGFR  stimulates cells to proliferate. Excessive  EGFR activation may...
TGF - β Signaling Pathway01:16

TGF - β Signaling Pathway

The TGF-β signaling pathway regulates cell growth, differentiation, adhesion, motility, and development. TGF-β ligands that induce TGF-β signaling are synthesized in their latent form. Several proteases or cell surface receptors such as integrins act upon the latent form, releasing the active ligand. There are three types of mammalian TGF-βs: (TGF-β1, TGF-β2, and TGF-β3) that bind as homodimers or heterodimers to TGF-β receptors. The TGF-β receptors are of three kinds RI, RII, and RIII. The RI...

Summary
This summary is machine-generated.

Transforming growth factor beta1 (TGF-beta1) enhances interferon-gamma (IFN-gamma) induced major histocompatibility complex class II (MHC-II) expression in mouse microglia at the transcriptional level. This effect differs in macrophages, where TGF-beta1 inhibits IFN-gamma induction.

Area of Science:

  • Immunology
  • Neuroscience
  • Molecular Biology

Background:

  • Interferon-gamma (IFN-gamma) is a key cytokine involved in immune responses.
  • Major histocompatibility complex class II (MHC-II) molecules present antigens to T helper cells.
  • Microglia are the primary immune cells of the central nervous system.

Purpose of the Study:

  • To investigate the combined effects of IFN-gamma and transforming growth factor beta1 (TGF-beta1) on MHC-II gene expression.
  • To determine if TGF-beta1 modulates IFN-gamma's effect on MHC-II expression in microglia and macrophages.
  • To elucidate the mechanism by which TGF-beta1 influences MHC-II expression.

Main Methods:

  • Primary mouse microglial and macrophage cultures were used.
  • MH-S macrophage cell line was utilized.
  • Cells were treated with IFN-gamma and TGF-beta1.
  • MHC-II gene expression was analyzed at the transcriptional and protein levels.
  • Reporter gene assays with MHC-II promoter constructs were performed.

Main Results:

  • IFN-gamma potently induced MHC-II gene expression in all cell types.
  • TGF-beta1 further elevated IFN-gamma-induced MHC-II expression in microglia at both gene and protein levels.
  • TGF-beta1 inhibited IFN-gamma-induced MHC-II expression in macrophages.
  • Reporter gene assays confirmed that TGF-beta1 enhances IFN-gamma's effect at the transcriptional level in microglia.

Conclusions:

  • TGF-beta1 differentially regulates IFN-gamma-induced MHC-II expression in microglia and macrophages.
  • TGF-beta1 acts transcriptionally to enhance MHC-II expression in microglia.
  • These findings highlight cell-specific roles of TGF-beta1 in modulating immune responses within the central nervous system.
