Background:

Antibody production requires B cell differentiation into plasma cells.
CD27 is a memory marker on B cells and a member of the tumor necrosis factor receptor (TNFR) family.

Purpose of the Study:

To investigate the role of CD27 signaling in B cell differentiation into plasma cells.
To identify key molecular players involved in this differentiation process.

Main Methods:

Purified peripheral blood B cells were stimulated with CD27 ligand (CD70) in the presence of various cytokines (IL-10, IL-2, IL-4, IL-6).
B cell differentiation into plasma cells was assessed.
CD40 signaling was evaluated for its effect on B cell pool expansion.

CD70 stimulation, in combination with IL-10, significantly increased plasma cell numbers.

This differentiation occurred specifically in CD27+ B cells.

IL-2 further enhanced IL-10 and CD70-induced plasma cell differentiation.

CD70 alone, or with IL-2, IL-4, or IL-6, did not promote plasma cell differentiation.

CD40 signaling promoted B cell pool expansion.

Conclusions:

CD27 ligand (CD70) is a critical factor for directing CD27+ memory B cell differentiation into plasma cells.
The cooperation between CD70 and IL-10 is essential for this process.
IL-2 can further enhance this differentiation pathway.
