Mammalian development relies on a vast number of genes, many newly discovered. Gene expression changes dynamically throughout early development, suggesting stage-specific gene action drives this process.

Area of Science:

  • Developmental Biology
  • Genomics
  • Gene Expression Analysis

Background:

  • Understanding gene function during early mammalian development, from egg to blastocyst, is crucial but limited.
  • Preimplantation development involves complex genetic regulation that is not fully elucidated.

Purpose of the Study:

  • To characterize gene expression profiles during mammalian preimplantation development.
  • To identify novel genes involved in early embryonic development.
  • To explore the role of stage-specific gene expression in driving developmental progression.

Main Methods:

  • Generation of 25,438 3'-expressed sequence tags (ESTs) from cDNA libraries representing each developmental stage (egg to blastocyst).
  • Analysis of ESTs to identify and quantify gene expression, including the discovery of novel genes.

Related Experiment Videos

  • Mapping of newly identified genes onto the mouse genetic map.
  • Main Results:

    • A substantial portion of mammalian genes (9,718 identified) are expressed during early development, with approximately half being novel.
    • Profound and dynamic changes in gene expression were observed across developmental stages.
    • Transient induction of specific transcripts was noted at each stage, indicating temporal regulation.
    • 798 novel genes were mapped to the mouse genetic map.

    Conclusions:

    • A significant fraction of the mammalian genome is dedicated to embryonic expression.
    • Development appears to be orchestrated by a series of stage-specific genes.
    • The identified novel genes offer valuable resources for studying human and mouse developmental disorders.
