Summary

Photosensitive poly(allylamine alpha-cyanocinnamylideneacetate) was used to create stronger alginate microcapsules. These light-cross-linked microcapsules effectively encapsulated and supported the proliferation of mouse leukemia cells.

Area of Science:

  • Polymer Chemistry
  • Materials Science
  • Biotechnology

Background:

  • Alginate microcapsules are widely used but can lack mechanical stability.
  • Developing methods to enhance microcapsule strength is crucial for various applications, including cell encapsulation.

Purpose of the Study:

  • To synthesize a photosensitive polymer, poly(allylamine alpha-cyanocinnamylideneacetate).
  • To fabricate and characterize alginate microcapsules coated with this photosensitive polymer.
  • To evaluate the mechanical strength and cell culture capabilities of the photo-cross-linked microcapsules.

Main Methods:

  • Coupling of an alpha-cyanocinnamylideneacetyl group to poly(allylamine) to create a photosensitive polymer.
  • Fabrication of alginate microcapsules followed by coating with the photosensitive polymer.

Related Experiment Videos

  • Light-induced cross-linking of the polymer coating to enhance microcapsule strength.
  • Assessment of microcapsule mechanical stability through agitation tests.
  • Evaluation of the permeability of the photo-cross-linked membrane to different proteins.
  • Entrapment and culture of IW32 mouse leukemia cells within the microcapsules.
  • Main Results:

    • The photosensitive polymer, poly(allylamine alpha-cyanocinnamylideneacetate), was successfully synthesized.
    • Microcapsules coated with this polymer exhibited significantly enhanced mechanical strength after light-induced cross-linking compared to unmodified poly(allylamine) coated capsules.
    • The photo-cross-linked membrane showed selective permeability to proteins (cytochrome C, myoglobin, serum albumin).
    • IW32 mouse leukemia cells encapsulated within the microcapsules proliferated to a high density (9 x 10(6) cells/ml) within 7 days.

    Conclusions:

    • Light-induced cross-linking of poly(allylamine alpha-cyanocinnamylideneacetate) provides an effective method for strengthening alginate microcapsules.
    • These enhanced microcapsules demonstrate potential for robust cell encapsulation and long-term cell culture applications.
