Dendritic cells (DCs) pulsed with thyroglobulin (Tg) can induce experimental autoimmune thyroiditis (EAT) in mice. This T cell-mediated autoimmune disease severity correlates with Tg concentration and Th1 cell activation.

Background:

  • Dendritic cells (DCs) are key antigen-presenting cells (APCs) that initiate and regulate immune responses.
  • Experimental autoimmune thyroiditis (EAT) is a T cell-mediated autoimmune disease and a model for human autoimmune thyroiditis.

Purpose of the Study:

  • To investigate the role of DCs in the induction of EAT.
  • To determine if DCs pulsed with thyroglobulin (Tg) can induce EAT.

Main Methods:

  • Naive DCs were purified from B10BR mice and pulsed with varying concentrations of porcine Tg (pTg).
  • pTg-pulsed DCs were transferred without adjuvant to syngenic BIOBR mice to induce EAT.
  • Thyroiditis severity, anti-pTg IgG2a antibodies, and cytokine levels (IFN-gamma, IL-4, IL-10) were assessed in recipient mice.
