    Background:

    • Fungal genomes contain repetitive DNA sequences, but their nature and distribution across major fungal groups remain incompletely understood.
    • Understanding the organization of repetitive DNA is crucial for fungal taxonomy and evolutionary studies.

    Purpose of the Study:

    • To investigate the characteristics of repeated DNA sequences in diverse fungal species.
    • To determine the abundance, size, and potential genomic location of repetitive DNA elements in fungi.

    Main Methods:

    • DNA:DNA reassociation studies were performed on several fungal species representing Basidiomycetes, Ascomycetes, and Phycomycetes.
    • Hydroxyapatite chromatography was utilized to analyze the reassociation kinetics of fungal DNA.
    • Thermal stability (T(e)50) of DNA duplexes was measured to assess sequence homology.

    Main Results:

    • All examined fungal species exhibited 10-20% repeated DNA sequences.
    • Approximately 100-110 copies of repeated DNA sequences, each around 4 x 10^7 daltons, were identified per genome.
    • Repeated DNA sequence homoduplexes displayed a T(e)50 difference of approximately 5°C compared to unfractionated whole DNA homoduplexes.

    Conclusions:

    • Repetitive DNA sequences are a common feature across major fungal phyla.
    • The observed characteristics suggest that repetitive DNA in fungi comprises both mitochondrial and nuclear DNA components.
    • Further research is warranted to fully elucidate the functional and evolutionary roles of these repetitive elements in fungal genomes.
