Background:

  • The thymus is traditionally recognized as the primary site for T cell differentiation.
  • The presence and function of B cells within the thymus are less understood.

Purpose of the Study:

  • To investigate the export of B cells from the thymus.
  • To characterize the developmental stages and origins of thymic emigrant B cells.

Main Methods:

  • Flow cytometry analysis of thymic emigrants from C57BL/6 mice.
  • Identification of B cell progenitors and mature B cells using specific markers (B220, IgM, CD19, VpreB, lambda5, pax-5).

Main Results:

  • The thymus exports approximately 3 x 10^4 mature B cells daily.
  • A significant portion (approx. 2 x 10^4/day) of these emigrant B cells develop intrathymically.
  • B cell progenitors (pro-B, pre-B) are present in the thymic cortex, with mature B cells located in deeper regions.

Conclusions:

  • The thymus contributes to the peripheral B cell pool, alongside its known role in T cell development.
  • The thymus actively exports intrathymically developed B cells.
