Murine double minute-2 (mdm2) mRNA expression is a significant prognostic indicator in soft tissue sarcoma (STS). Lower mdm2 mRNA levels (<50 zmol/amol) are linked to reduced survival, highlighting its clinical relevance.

Background:

  • Overexpression of murine double minute-2 (mdm2) protein, often interacting with tumor suppressor p53, has known oncogenic properties.
  • The clinical significance of mdm2 mRNA expression, however, remains under-investigated in cancer prognosis.

Purpose of the Study:

  • To investigate the clinical relevance of mdm2 mRNA expression as a prognostic factor in soft tissue sarcoma (STS).

Main Methods:

  • Quantitative reverse transcription polymerase chain reaction (RT-PCR) was used to analyze mdm2 mRNA expression in 65 STS samples.
  • Mdm2 mRNA levels were normalized to glyceraldehyde-3-phosphate dehydrogenase (GAPDH) expression.

Main Results:

  • Patients with an mdm2/GAPDH mRNA ratio below 50 zmol/amol exhibited significantly reduced survival compared to those with a ratio of >=50 (p=0.0241).

  • Multivariate analysis confirmed mdm2 mRNA expression as an independent prognostic factor, with ratios <50 associated with a 5.6-fold increased risk in stages 2 and 3 (p=0.0041).
