Summary

Galanin, but not its fragment, antagonizes central alpha(2)-adrenoceptor effects on blood pressure and heart rate. This interaction within the nucleus tractus solitarii suggests a reciprocal modulatory mechanism between galanin and alpha(2)-receptors.

Background:

  • Alpha(2)-adrenoceptors centrally regulate cardiovascular function.
  • Galanin is a neuropeptide with complex physiological roles.
  • The interaction between galanin and alpha(2)-adrenoceptors in cardiovascular control is not fully understood.

Purpose of the Study:

  • To investigate the modulatory effects of galanin and its N-terminal fragment on central alpha(2)-adrenoceptor-mediated cardiovascular responses.
  • To explore the reciprocal interaction between galanin and alpha(2)-adrenoceptors in the nucleus tractus solitarii (NTS).

Main Methods:

  • Quantitative receptor autoradiography to assess binding affinities (K(d), B(max)) of radioligands.
  • Cardiovascular analysis in vivo following intracisternal coinjections of agonists and peptides.

    • Galanin, coinjected with clonidine (a selective alpha(2)-receptor agonist), induced vasopressor and tachycardic responses, counteracting clonidine's hypotensive effect.
    • Galanin (1 nM) increased K(d) and B(max) of [(3)H]p-Aminoclonidine binding in the NTS, suggesting antagonism at alpha(2)-adrenoceptors.
    • Clonidine increased galanin binding in the NTS, indicating a reciprocal facilitatory effect on galanin receptors.

    Conclusions:

    • Galanin exerts an antagonistic effect on central alpha(2)-adrenoceptor-mediated cardiovascular responses, while galanin-(1-15) does not.
    • Alpha(2)-adrenoceptors reciprocally facilitate galanin binding.
    • These findings suggest a functional and reciprocal interaction between galanin and alpha(2)-receptors within the NTS regulating cardiovascular control.
