Background:

  • Heparin affinity chromatography was used to study the interaction of mouse brain proteoglycans with heparin.
  • Recombinant C-terminal fragments of neurocan, with and without chondroitin sulfate chains, were produced and analyzed.

  • Neurocan, unlike brevican, demonstrated retention on a heparin affinity matrix.
  • A recombinant C-terminal fragment of neurocan also bound to heparin, independent of its chondroitin sulfate substitution at physiological NaCl concentrations.

Conclusions:

  • Neurocan possesses intrinsic heparin-binding capabilities mediated by its C-terminal domain.
  • This heparin binding may facilitate interactions with other heparin-binding molecules and direct neurocan secretion in polarized cells.
