Summary

Iron overload damages kidney lysosomes, making them more fragile and dense. These iron-induced changes in kidney lysosomes can be reversed by reducing the iron load.

Area of Science:

  • Nephrology
  • Cell Biology
  • Toxicology

Background:

  • Iron overload is linked to organ damage, including the liver.
  • The specific impact of iron overload on kidney lysosome pathophysiology requires further investigation.

Purpose of the Study:

  • To investigate the effects of experimentally induced iron overload on the pathophysiology of kidney lysosomes in male rats.

Main Methods:

  • Iron overload was induced in rats using intraperitoneal injections of Fe-dextran.
  • Kidney homogenates were analyzed using differential and density gradient centrifugation.
  • Kidney iron load and lysosomal changes were assessed at various time points.

Main Results:

  • Fe-dextran administration significantly increased kidney iron load.
  • Iron primarily accumulated in lysosomes, altering their behavior.
  • Lysosomes exhibited increased fragility and density, correlating with iron accumulation levels and duration.

Conclusions:

  • Iron overload induces distinct pathological changes in kidney lysosomes.
  • These iron-induced lysosomal alterations are dose- and time-dependent.
  • Reducing iron load can reverse the observed changes in kidney lysosomes.
