Background:

  • T-cell receptor (TCR) binding to antigen (Ag) leads to phosphorylation, internalization, and degradation.
  • The dynamic changes in TCR mRNA levels following antigen engagement are not fully understood.

Purpose of the Study:

  • To investigate whether TCR binding to antigen is accompanied by a specific increase in TCR mRNA levels.
  • To determine the relationship between antigen stimulation and TCR gene transcription.

Main Methods:

  • Peripheral blood mononuclear cells (PBMC) and a T-cell clone were cultured with superantigens and an alloantigen.
  • Quantitative analysis of TCR beta-chain variable (TCRBV)-specific mRNA levels.
  • Reporter gene assays using cloned TCRBV gene promoters.
