Thymidine phosphorylase (dThdPase) expression in gastric cancer correlates with increased microvessels and reduced apoptosis. This suggests dThdPase may promote tumor growth and indicate a poorer prognosis in these cancers.

Area of Science:

  • Oncology
  • Molecular Biology
  • Cancer Research

Background:

  • Thymidine phosphorylase (dThdPase), also known as platelet-derived endothelial cell growth factor (PD-ECGF), is upregulated in various human carcinomas.
  • Elevated dThdPase expression is linked to increased intratumoral microvessel density (IMVD) and unfavorable patient prognosis.

Purpose of the Study:

  • To investigate the role of dThdPase in apoptosis, IMVD, and p53 expression in human gastric carcinomas.
  • To determine the association between dThdPase expression levels and key indicators of tumor progression.

Main Methods:

  • dThdPase expression was assessed in 34 early and 36 advanced gastric carcinomas.
  • Intratumoral microvessel density (IMVD) and apoptotic index (AI) were quantified in dThdPase-positive and dThdPase-negative tumor areas.

Related Experiment Videos

  • Statistical analyses were performed to compare IMVD and AI between groups and to assess correlations.
  • Main Results:

    • dThdPase expression was detected in 35.3% of early and 55.6% of advanced gastric carcinomas.
    • Mean IMVD was significantly higher in dThdPase-positive tumors compared to dThdPase-negative tumors (P<0.05).
    • Mean AI was significantly higher in dThdPase-negative tumors than in dThdPase-positive tumors (P<0.05), with an inverse correlation between AI and IMVD (P<0.001).

    Conclusions:

    • dThdPase expression in gastric carcinomas is associated with increased IMVD and decreased apoptosis.
    • These findings suggest that dThdPase contributes to angiogenesis and inhibits apoptosis, potentially driving gastric cancer progression.
