Area of Science:

  • Neurology
  • Pharmacology
  • Geriatrics

Background:

  • Alzheimer's disease (AD) is characterized by cognitive decline, impacting daily living, behavior, and caregivers.
  • Clinically meaningful outcomes like the Disability Assessment for Dementia (DAD) and Neuropsychiatric Inventory (NPI) are crucial for assessing AD treatments.
  • Galantamine, a novel AD treatment, offers a dual mechanism of action: nicotinic receptor modulation and acetylcholinesterase inhibition.

Purpose of the Study:

  • To review the efficacy of galantamine in improving daily functioning and behavioral disturbances in Alzheimer's disease patients.
  • To assess the impact of galantamine on caregiver burden and sleep quality.
  • To highlight galantamine's sustained benefits in managing Alzheimer's disease.

Main Methods:

  • Review of clinical studies evaluating galantamine in Alzheimer's disease patients.
  • Assessment of outcomes using scales such as Disability Assessment for Dementia (DAD), Neuropsychiatric Inventory (NPI), and Pittsburgh Sleep Quality Index (PSQI).
  • Comparison of galantamine treatment groups with placebo groups regarding patient function, behavior, and caregiver time.

Main Results:

  • Galantamine treatment led to significantly better outcomes in activities of daily living (ADL) compared to placebo over 5 months.
  • Twelve months of galantamine treatment preserved patients' functional abilities.
  • Galantamine significantly improved behavioral disturbances and reduced caregiver time spent assisting patients, without negatively impacting sleep quality.

Conclusions:

  • Galantamine demonstrates positive and sustained effects on patient behavior and daily functioning in Alzheimer's disease.
  • The benefits of galantamine extend to reducing the burden on caregivers.
  • Galantamine offers a favorable safety profile, particularly regarding sleep quality, compared to other cholinergic treatments.
