Summary

Gold nanoparticles enhance radiation dose and cell killing in vitro and in vivo when irradiated with kilovoltage X-rays. This dose enhancement effect shows promise for improving cancer radiotherapy, particularly with low-energy photon sources.

Area of Science:

  • Medical Physics
  • Radiation Oncology
  • Nanotechnology

Background:

  • Gold nanoparticles (GNPs) are being explored for their potential in cancer therapy.
  • High-Z materials can enhance radiation dose deposition through secondary electron emission.

Purpose of the Study:

  • To investigate dose enhancement and radiosensitization using gold particles with low-energy photons.
  • To evaluate the efficacy of gold particles in cell cultures and in vivo tumor models.

Main Methods:

  • Cell survival assays (clonogenic) were performed on CHO-K1, EMT-6, and DU-145 cells irradiated with kilovoltage X-rays and Cs-137 gamma rays in the presence of gold particles.
  • In vivo studies involved injecting gold particles into EMT-6 tumors in scid mice, followed by irradiation and assessment of tumor cell killing.

Related Experiment Videos

Main Results:

  • Dose enhancement was confirmed with kilovoltage X-rays using Fricke dosimetry and cell killing, yielding dose modification factors (DMF) around 1.43.
  • Gold particles did not show significant dose enhancement with Cs-137 gamma rays.
  • In vivo studies demonstrated modest radiosensitization but highlighted challenges with heterogeneous gold particle distribution.

Conclusions:

  • Gold microspheres can increase the biologically effective dose when cells or tumors are exposed to kilovoltage photon beams.
  • High-Z particles like gold may improve the therapeutic ratio in radiotherapy, especially with low-energy photon brachytherapy.
