Purpose of the Study:

  • To investigate the association between IL-10 production and joint destruction in inflammatory arthritis.
  • To determine if genetic polymorphisms in the IL-10 gene influence the extent of joint damage.

Main Methods:

  • Compared IL-10 mRNA levels in synovial biopsies from patients with destructive versus non-destructive joint disease.
  • Analyzed IL-10 genotypes and their correlation with radiographic joint damage progression over 12 years in female rheumatoid arthritis patients.
  • Assessed IL-10 production in stimulated whole blood cultures from donors with different IL-10 alleles.

Main Results:

  • Significantly lower IL-10 mRNA levels were found in synovial biopsies from patients with erosive (destructive) joints compared to non-erosive joints (P < 0.03).

Conclusions:

  • Both synovial biopsy and immunogenetic data suggest a strong association between variations in IL-10 production and the degree of joint destruction in inflammatory arthritis.
  • Genetic factors influencing IL-10 production play a role in determining the severity of joint damage in rheumatoid arthritis.
