Purpose of the Study:

  • To determine the composition, bacteriostatic, and radical scavenging potential of propolis samples.
  • To differentiate between fresh and aged propolis based on chemical and biological properties.
  • To evaluate the utility of radical scavenging potential as a quality indicator for propolis.

Main Methods:

  • Analysis of fifteen propolis samples from diverse origins.
  • Quantification of polyphenols and flavonoids.
  • Determination of bacteriostatic activity against Bacillus subtilis, Staphylococcus aureus, and Escherichia coli.
  • Assessment of radical scavenging potential (ROO*).
  • Statistical analysis (variance analysis) to identify significant differences.

Main Results:

  • Significant differences in polyphenol and flavonoid content between fresh and aged propolis (p < 0.05).
  • Fresh propolis required 80 µg/ml for B. subtilis/S. aureus inhibition and 800 µg/ml for E. coli inhibition.
  • Aged propolis required 100 µg/ml for B. subtilis/S. aureus inhibition and 1000 µg/ml for E. coli inhibition.
  • Fresh propolis showed a minimum flavonoid content of 18 g/100 g and max ROO* scavenging of 4.3 µg/ml.
  • Aged propolis had ~20% lower flavonoids but higher ROO* scavenging (5.7–6.4 µg/ml).

Conclusions:

  • Propolis state (fresh or aged) can be distinguished by flavonoid profiles and biological activities.
  • Bacteriostatic and radical scavenging activities differ between fresh and aged propolis.
  • Radical scavenging potential shows promise as an objective measure for propolis quality assessment.
