Summary

The WHSC2 gene, linked to Wolf-Hirschhorn syndrome, is found in endothelial cells and may act as a transcriptional repressor. HIV-1 Tat downregulates WHSC2, suggesting a role in viral-host interactions.

Area of Science:

  • Genetics
  • Molecular Biology
  • Cell Biology

Background:

  • The WHSC2 gene, located in the critical region of Wolf-Hirschhorn syndrome, is identified in endothelial cells.
  • WHSC2 expression is modulated by HIV-1 Tat but not by angiogenic or pro-differentiative factors.

Purpose of the Study:

  • To characterize the WHSC2 gene and its protein product.
  • To investigate the cellular localization and potential function of WHSC2.

Main Methods:

  • In vitro translation to determine protein molecular weight.
  • Transfection of NIH-3T3 cells with whsc2-GFP to visualize nuclear translocation.
  • Bioinformatic analysis to identify protein motifs (e.g., helix-loop-helix).

Main Results:

Related Experiment Videos

  • WHSC2 encodes a 57 kDa basic polypeptide.
  • The protein possesses two nuclear translocation sequences, mediating active transport to the nucleus.
  • A helix-loop-helix (HLH) motif was identified in region 130-185 of WHSC2.

Conclusions:

  • WHSC2 is actively transported to the nucleus.
  • The presence of an HLH motif suggests WHSC2 may function as a transcriptional repressor, potentially influencing cell differentiation and cell cycle progression.
