Summary

Cytokine profiles in autoimmune thyroiditis-prone rats show a shift towards IL-10 over IFNgamma. This suggests a potential role for IL-10 in the development of spontaneous disease.

Background:

  • Cytokines play a critical role in modulating autoimmune disease progression.
  • The specific role of cytokine kinetics in the spontaneous development of autoimmune thyroiditis remains incompletely understood.
  • This study investigates cytokine production in rats genetically predisposed to autoimmune thyroiditis.

Purpose of the Study:

  • To compare the cytokine production profiles of T cells from autoimmune thyroiditis-prone rats (NB line BB/Wor) and control rats (Wistar) after activation.
  • To elucidate the role of antigen-presenting cells (APCs) in modulating T cell responses and cytokine production.
  • To examine the presence and localization of key cytokines within the thyroid tissue.

Main Methods:

  • T cell splenocytes from NB and Wistar rats were activated with thyroglobulin (Tg) or Concanavalin A (Con A) in the presence of homologous or heterologous irradiated splenocytes as APCs.
  • Cytokine concentrations (IL-2, IL-4, IL-10, TNF-alpha, IFN-gamma) and mRNA expression were measured using ELISA and RT-PCR.
  • Immunohistochemistry was employed to detect IL-10 in thyroid tissues.
  • T cell proliferation was assessed via 3H thymidine uptake.

Main Results:

  • NB rat T cell cultures exhibited higher concentrations of IL-10 compared to IFN-gamma following activation, whereas Wistar rat cultures showed the reverse.
  • Wistar APCs significantly enhanced T cell proliferation and cytokine responses in NB rats.
  • Thyroid tissues from NB rats contained IL-10, but not IFN-gamma, while control rat thyroids lacked IL-10.

Conclusions:

  • Splenocyte responses in LT-prone BB/Wor rats are characterized by a predominant IL-10 production.
  • These findings highlight a potential role for IL-10 in the pathogenesis of spontaneous autoimmune thyroiditis.
  • Further research is warranted to investigate the source and function of intrathyroidal IL-10 in this model.
