Purpose of the Study:

  • To prospectively evaluate the impact of low-dose estradiol on arterial endothelial and smooth muscle function in healthy men.
  • To investigate the dose-dependent effects of estradiol on vascular reactivity.

Main Methods:

  • Twenty-three healthy men were randomized into three groups receiving testosterone with varying doses of estradiol (0 mg, 10 mg, or 20 mg).
  • Vascular reactivity was assessed using high-resolution ultrasound to measure brachial artery diameter changes during reactive hyperemia (flow-mediated dilation, FMD) and after nitroglycerin administration.

  • Main Results:

    • Estradiol administration resulted in a dose-dependent increase in plasma estradiol levels.
    • Flow-mediated dilation (FMD), an indicator of endothelial function, showed a significant dose-dependent improvement with estradiol treatment at 6 months (P=0.02).
    • The increase in estradiol levels correlated positively with improvements in FMD (P=0.01), while nitroglycerin responses remained unchanged, indicating specific effects on endothelial function.

    • Low-dose estradiol supplementation enhances arterial endothelial function in healthy young men.
    • Estradiol may play a role in maintaining vascular health in men.
