Nitric oxide (NO), an inorganic gas, acts as a potent second messenger in most animal and plant tissues. NO diffuses out of the cells that produce it and enters the neighboring cells to generate a downstream response. Summary

Nitric oxide synthases (NOS) are enzymes involved in gastric acid secretion. This study found neuronal NOS (nNOS) expressed in rat parietal cells, suggesting its role in regulating stomach acid production.

Background:

  • Nitric oxide synthases (NOS) produce nitric oxide (NO), a signaling molecule.
  • NO is implicated in regulating gastric acid secretion.
  • The specific NOS isoform in gastric oxyntic mucosa is not well-defined.

Purpose of the Study:

  • To determine the cellular localization of NOS in rat oxyntic mucosa.
  • To identify the specific NOS isoform expressed in parietal cells.
  • To investigate the role of NOS in gastric acid secretion regulation.

Main Methods:

  • NADPH-diaphorase histochemistry on rat oxyntic mucosa.
  • Immunohistochemistry using isoform-specific NOS antibodies.
  • Western blot analysis and RT-PCR on isolated rat parietal cells.

Main Results:

  • NADPH-diaphorase and nNOS immunoreactivity were localized to parietal cells in the oxyntic mucosa.
