    Area of Science:

    • Medical Genetics
    • Skeletal Dysplasias
    • Rare Diseases

    Background:

    • Pycnodysostosis is a rare inherited disorder.
    • It is inherited as an autosomal recessive trait.
    • Characterized by specific skeletal abnormalities.

    Purpose of the Study:

    • To describe the clinical and radiological features of pycnodysostosis.
    • To differentiate pycnodysostosis from similar bone disorders.
    • To highlight the generally favorable prognosis.

    Main Methods:

    • Case report of two patients with pycnodysostosis.
    • Review of clinical and radiological findings.
    • Comparison with existing literature on osteosclerosis disorders.

    Main Results:

    • Generalized osteosclerosis (medium degree).
    • Persistent open sutures and fontanels into adulthood.
    • Mandibular hypoplasia, frontal/occipital prominence, acro-osteolysis, and bone fragility.
    • Disproportionate dwarfism is common.

    Conclusions:

    • Pycnodysostosis is a distinct autosomal recessive osteosclerosis syndrome.
    • Key features include specific craniofacial and skeletal anomalies.
    • Prognosis is favorable, contrasting with osteopetrosis.
