Summary

Endotoxin-induced metabolic stress in rats reduces lymphocyte glutamine utilization and proliferation. This decrease in glutamine use by lymphocytes is linked to lower glutaminase activity, impacting immune cell function.

Area of Science:

  • Immunology
  • Metabolic Biochemistry

Background:

  • Supplemental glutamine may benefit lymphocyte function during metabolic stress.
  • The precise mechanisms of glutamine utilization by stressed lymphocytes remain unclear.
  • This study investigates glutamine use and proliferation in lymphocytes under endotoxin-induced stress.

Purpose of the Study:

  • To determine the effect of endotoxin-induced stress on in vitro glutamine utilization by lymphocytes.
  • To assess the impact of stress on glutamine-dependent proliferation of activated lymphocytes.
  • To explore the relationship between glutamine metabolism and lymphocyte function post-stress.

Main Methods:

  • Metabolic stress was induced in rats via intraperitoneal endotoxin injection.
  • Lymphocytes were isolated at various time points (6, 12, 24, 48 hours) post-injection.

Related Experiment Videos

  • Lymphocyte proliferation, glutamine utilization rate, and glutaminase activity were measured in vitro.
  • Main Results:

    • Lymphocyte proliferation was suppressed 12, 24, and 48 hours after endotoxin administration.
    • The glutamine concentration required for maximal lymphocyte response decreased over time post-stress.
    • Both glutamine utilization rate and glutaminase activity in activated lymphocytes showed a time-dependent decrease.

    Conclusions:

    • Glutamine utilization by lymphocytes is significantly reduced in the late phase after endotoxin injection.
    • Decreased glutaminase activity contributes to the reduced glutamine utilization.
    • Impaired glutamine metabolism is associated with suppressed proliferation of mitogen-activated lymphocytes.
