Background:

  • Osteoarthritis (OA) is a prevalent degenerative joint disease causing significant disability and reduced quality of life.
  • Current treatment options for OA often have limitations in efficacy and safety.

Purpose of the Study:

  • To systematically review randomized controlled trials (RCTs) assessing the efficacy and safety of glucosamine for osteoarthritis (OA).

Main Methods:

  • Comprehensive literature search of MEDLINE, Embase, Current Contents, and Cochrane Controlled Trials Register up to November 1999.
  • Inclusion criteria specified RCTs evaluating glucosamine in OA, including placebo-controlled and comparative studies, both single and double-blinded.
  • Data abstraction and quality assessment (Jadad 1995) were performed independently; continuous and dichotomous outcomes were pooled using standardized mean differences and Peto Odds Ratios, respectively.

  • Sixteen RCTs were identified, collectively indicating glucosamine is effective and safe for OA.
  • In 13 placebo-controlled RCTs, glucosamine demonstrated superiority in all but one study.
