Area of Science:

  • Pharmacology
  • Endocrinology
  • Oncology

Background:

  • Selective estrogen receptor modulators (SERMs) offer targeted estrogenic effects.
  • Osteoporosis and breast cancer are significant health concerns for postmenopausal women.

Purpose of the Study:

  • To evaluate the efficacy of raloxifene in reducing fracture risk and breast cancer incidence.
  • To assess the impact of raloxifene on cardiovascular risk factors and safety profile.

Main Methods:

  • Clinical trials in humans and animal studies were conducted.
  • The 60 mg dose of raloxifene was administered and outcomes were monitored.

Main Results:

  • Raloxifene demonstrated a 30%-40% reduction in spine fractures and a 60%-70% reduction in breast cancer risk.
  • A decrease in ankle fractures was observed, but not hip or wrist fractures.
  • Raloxifene lowered total and LDL cholesterol, with no increase in triglycerides.

Conclusions:

  • Raloxifene is effective in reducing spine fractures and breast cancer risk in postmenopausal women.
  • While offering cardiovascular benefits like cholesterol reduction, it does not prevent major cardiovascular events.
  • Potential side effects include an increased risk of thromboembolic disease, leg cramps, and hot flushes.
