    Summary

    Halothane anesthesia significantly elevated serum bromide levels in healthy volunteers to psychoactive concentrations. These elevated bromide levels may explain prolonged mood and cognitive changes observed after halothane exposure.

    Background:

    • Halothane is a widely used inhalation anesthetic.
    • Prolonged mood and cognitive changes have been anecdotally reported after halothane anesthesia.
    • The underlying biochemical mechanisms for these changes remain unclear.

    Purpose of the Study:

    • To investigate the impact of halothane anesthesia on serum bromide concentrations.
    • To determine if elevated bromide levels correlate with psychoactive effects.

    Main Methods:

    • Seven healthy male volunteers underwent halothane-oxygen anesthesia without surgical procedures.
    • Serum bromide concentrations were measured before anesthesia, on the second day, and on the ninth day after anesthesia.
    • Plasma and urinary fluoride levels were also monitored.

    • Serum bromide concentrations increased significantly from baseline (0.6 mEq/l) to 2.9 mEq/l on day two and remained elevated at 2.5 mEq/l on day nine.
    • The observed bromide levels are considered psychoactive.
    • No significant increase in plasma or urinary fluoride concentrations was detected.

    Conclusions:

    • Halothane anesthesia leads to a sustained increase in serum bromide concentrations to psychoactive levels.
    • Elevated bromide may be responsible for the previously observed post-halothane mood and cognitive disturbances.
    • Further research is warranted to fully elucidate the neurotoxicological effects of halothane-induced hyperbromemia.
