Receptor-mediated Endocytosis

8.1K
Receptor-mediated endocytosis is when bulk amounts of specific molecules are imported into a cell after binding to cell surface receptors. The molecules bound to these receptors are taken into the cell through inward folding of the cell surface membrane, which is eventually pinched off into a vesicle within the cell. Structural proteins, such as clathrin, coat the budding vesicle.

Endocytosis01:16

Endocytosis

12.9K
Eukaryotic cells acquire nutrients for growth and proliferation. Nutrients and other molecules that require degradation are internalized from the extracellular space by a process called endocytosis. The term ‘endocytosis' was first coined by Christian de Duve in 1963.
Endocytosis always begins with the plasma membrane enclosing an incoming molecule to form a transport vesicle which, in some cases, can be coated with a protein called ‘clathrin.' Endocytosed material is either...

The Early Endosome: Endocytosis of Transferrin

4.9K
Essential proteins such as insulin or low-density lipoprotein (LDL) and micronutrients such as iron enter a eukaryotic cell through receptor-mediated endocytosis. Subsequently, the early endosomes fuse with the vesicles containing such receptor-ligand complexes and play a vital role in sorting the incoming ligands and receptors. While the ligands are either degraded inside the vesicle or released into the cytosol, their receptors are returned to the plasma membrane for further rounds of...

The Eukaryotic Promoter Region02:40

The Eukaryotic Promoter Region

19.0K
The eukaryotic promoter region is a segment of DNA located upstream of a gene. It contains an RNA polymerase binding site, a transcription start site, and several cis-regulatory sequences.  The proximal promoter region is located in the vicinity of the gene and has cis-regulatory sequences and the core promoter. The core promoter is the binding site for RNA polymerase and is usually located between -35 and +35 nucleotides from the transcription start site. The distal promoter regions are...

5-Number Summary01:04

5-Number Summary

5.7K
In a dataset, the 5-number summary includes the minimum data value, the data value of the first quartile, the median data value or data value of the second quartile, the data value of the third quartile, and the maximum data value. These 5 data values can be visualized as a box and whisker plot.
In a box plot, the minimum and maximum data values represent the lower and upper whiskers in the graph, and the median is designated as the center of the box in the chart. The first quartile and third...

Area of Science:

  • Cell Biology
  • Molecular Biology
  • Protein Interactions

Background:

  • Rabphilin is a secretory vesicle protein interacting with Rab3.
  • Endocytosis is a crucial cellular process for internalization of molecules.

Purpose of the Study:

  • Investigate Rabphilin's role in endocytosis.
  • Determine the influence of Rabphilin-Rab3 interaction on endocytosis.

Main Methods:

  • Utilized point mutants of Rabphilin (V61A, L83A) in HIT-T15 cells.
  • Assessed transferrin endocytosis rates.
  • Analyzed protein interactions and cellular localization.

Main Results:

  • Rabphilin V61A mutant enhanced transferrin internalization.
  • Rabphilin L83A mutant, unable to interact with Rabaptin-5, did not enhance endocytosis.
  • Rabphilin's action is negatively modulated by Rab3.

Conclusions:

  • Rabphilin promotes receptor-mediated endocytosis.
  • Rab3 negatively regulates Rabphilin's pro-endocytic function.
  • Rabphilin facilitates rapid recycling of secretory vesicle components.
