Background:

  • Fusarium oxysporum f. sp. lycopersici causes vascular wilt in tomato plants.
  • Pectinolytic enzymes secreted by F. oxysporum may aid in host plant penetration and colonization.

Purpose of the Study:

  • To isolate and characterize a novel pectinolytic enzyme gene, pg5, from F. oxysporum.
  • To investigate the role of pg5 in the pathogenesis of tomato vascular wilt.

Main Methods:

  • Isolation and cloning of the pg5 gene.
  • Heterologous expression of pg5 in a bacterial system.
  • Reverse transcription-PCR to analyze pg5 expression during infection.

Main Results:

  • A novel extracellular endopolygalacturonase (endoPG) gene, pg5, was identified and found to be conserved across F. oxysporum formae speciales.
  • The pg5 gene product exhibits endoPG activity and its transcript accumulation is regulated by pectin and D-galacturonic acid.
  • pg5 is expressed in tomato roots during early infection stages, but its inactivation did not impact virulence.

Conclusions:

  • pg5 encodes a conserved endoPG in F. oxysporum with pectin-regulated expression.
  • Despite early expression during infection, pg5 is not essential for the virulence of F. oxysporum on tomato plants.
