Background:

  • Acetaldehyde is the primary ethanol metabolite.
  • Its role in ethanol's reinforcing effects is debated; brain acetaldehyde may be reinforcing, while blood acetaldehyde is considered aversive.

Purpose of the Study:

  • To investigate the reinforcing and/or aversive effects of acetaldehyde and ethanol.
  • To determine if acetaldehyde contributes to the reinforcing properties of alcohol.

Main Methods:

  • Wistar rats underwent a conditioned reinforcement procedure.
  • An olfactory stimulus was paired with intraperitoneal injections of ethanol (0-2 g/kg) or acetaldehyde (0-150 mg/kg).
  • Stimulus preference or aversion was assessed post-conditioning.

  • Ethanol doses of 1.0 and 2.0 g/kg induced significant stimulus aversions.
  • Acetaldehyde induced a biphasic stimulus preference, peaking around 20 mg/kg.
  • No aversive effects were observed with increasing acetaldehyde doses.

  • Systemic acetaldehyde injections produced significant stimulus preferences in rats.
  • These findings suggest acetaldehyde may mediate alcohol's reinforcing effects.
