Intercellular adhesion molecule 1 (ICAM-1) gene G/R 241 polymorphism is linked to Behçet

Area of Science:

  • Immunogenetics
  • Vascular Biology
  • Rheumatology

Background:

  • Intercellular adhesion molecule 1 (ICAM-1) is expressed in vascular endothelial cells and inflammatory infiltrates in Behçet's disease (BD) lesions.
  • ICAM-1 gene polymorphisms may influence inflammatory processes contributing to vascular damage in BD.

Purpose of the Study:

  • To investigate the association between ICAM-1 gene polymorphisms and susceptibility to Behçet's disease (BD).

Main Methods:

  • Genotyping of 74 Italian BD patients and 228 healthy controls for ICAM-1 polymorphisms at codons 241 and 469 using polymerase chain reaction and allele-specific oligonucleotide techniques.
  • Statistical analysis to compare allele and genotype frequencies between patients and controls.

Main Results:

Related Experiment Videos

  • The G/R 241 polymorphism was significantly more frequent in BD patients (20.3%) compared to controls (5.7%) (p = 0.001, OR 4.2).
  • No significant difference was observed for the E/K 469 genotype distribution between groups.
  • A trend towards higher R241 frequency was noted in BD patients with articular manifestations.

Conclusions:

  • The G/R 241 polymorphism of the ICAM-1 gene is associated with an increased susceptibility to Behçet's disease.
  • This finding suggests a role for ICAM-1 genetic variations in the pathogenesis of BD.
