Summary

Fibroblast growth factors (FGFs) 3 and 8 are crucial for zebrafish ear development. Disrupting either FGF3 or FGF8 impairs otic vesicle formation, with combined disruption severely affecting otic tissue and early development.

Area of Science:

  • Developmental Biology
  • Genetics
  • Molecular Biology

Background:

  • Fibroblast growth factors (FGFs) are vital signaling molecules regulating embryonic development.
  • FGF signaling plays a role in the induction of the otic placode, a precursor to the inner ear.
  • Specific FGFs, fgf3 and fgf8, have been implicated in vertebrate otic development.

Purpose of the Study:

  • To functionally analyze the roles of fgf3 and fgf8 in zebrafish otic development.
  • To investigate the cooperative roles of fgf3 and fgf8 in otic placode induction.
  • To understand how alterations in fgf3 and fgf8 expression impact early ear formation.

Main Methods:

  • Analysis of acerebellar (ace) mutants to assess fgf8 function.
  • Injection of antisense morpholino oligomers (MO) to disrupt fgf3 translation.

Related Experiment Videos

  • Expression analysis of otic markers pax8 and pax2.1.
  • Examination of fgf3 and fgf8 expression patterns during gastrulation.
  • Investigating the effects of one-eyed pinhead (oep) mutations and retinoic acid treatment.
  • Main Results:

    • Disruption of either fgf3 or fgf8 individually caused a moderate reduction in otic vesicle size.
    • Combined disruption of fgf3 and fgf8 led to severe reduction or complete loss of otic tissue.
    • fgf3 and fgf8 are expressed in specific patterns during gastrulation, including the germring and rhombomere 4.
    • Altered expression patterns of fgf3 and fgf8 corresponded to changes in otic induction.
    • Loss of oep function delayed otic induction by ablating mesendodermal FGF sources.
    • Retinoic acid treatment expanded fgf3, fgf8, and pax8 expression, inducing supernumerary and ectopic otic vesicles.

    Conclusions:

    • FGF3 and FGF8 cooperate during gastrulation to induce otic placode differentiation in zebrafish.
    • These FGFs are essential for both early otic induction and subsequent otic vesicle development.
    • The precise spatial and temporal expression of fgf3 and fgf8 is critical for normal ear formation.
