Summary

Scanning electron microscopy (SEM) of rabbit aorta endothelium is most reliable when aortas are fixed in situ at physiological pressure. This method preserves endothelial integrity, unlike ex vivo fixation which causes sub-endothelial contraction and difficult interpretation.
